Message ISO is basically a gain on every pixel on the cameras CCD sensor. The highest you set it, the grainier the pictures will look as every little pixel will be artifically gained (and the gain is not neceessary the true color that spot has in real life)

As far as F-stops go, the lower the number, the smaller focal depth you get. So if you have something like 2.8, most of the lens radius is uncovered, but youll be only able to keep maybe 3cm of depth in focus. The stuff before it and after it will look blurry. Oh yeah anything over 1/60 shutter speed should be supported on a tripod, otherwise youll get progressively more blur
